How to make a Gel? (for electrophoresis)


 Hello everyone
Hope you have done well in the exam,

So i suppose everyone knows about DNA Gel electrophoresis, and today
i am gonna show you all how to actually make the gel out of powder and water~




you will need a Agarose Gel powder 1st

Some concentration TE buffer, but we use TAE most of the time
which is TE + acetic acid
It is used to stabilized to DNA and prevent degradation

a 100mL measuring cyclinder

and lastly the gel tank, basically a container.

now you need to weight the gel~
for 1% gel, 1g of agarose powder will do

weighting~that's not my hand

and mix 10ml of Concentration buffer with 90ml of water

then just pour the solution into the powder, and try to swirl the beaker to dissolve the powder

and you sent it into a microwave oven

heat for 30 seconds at high power!!



it should be quite warm after the 1st heating, and it is recommend to 
swirll it for a while since microwave does not mixed the solution well

after a few heating cycles, a clear solution should be obtained
it should be almost boiling by now.

then you cool the solution by running it under a cool tap water, or just put into a 60 C water bath for half an hour.

finally, pour the agar into the tank~make sure you don't create bubble
they does not look nice on agar.
 I am sure you don't want pimple on your face, same theory applies


Don't move or play with the tank after you pour in the solution!!!!!!!!!!!!!!
it will result into a rough surface, which also not really that nice to see
and might affect the result later on during DNA analysis

finally, you load in the DNA into the wells, and you run in this matter~~~~

MIliQ water-water so pure you cannot drink!!



You need a ultra pure water but don't know how to purify?
MiliQ water is the answer!!!!!!

Milli-Q refers to water that has been purified and deionized to a high degree (typically 18.2 MΩ·cm) by a water purification systems manufactured by Millipore Corporation. It is also a registered trademark of Millipore. The term is also used to refer to water produced by any similar system.

Milli-Q water uses resin filters and deionization to purify the water. The system monitors the ion concentration by measuring the electrical resistivity of the water. Higher resistivity means fewer charge-carrying ions. Most Milli-Q systems dispense the water through a 0.22 μm membrane filter.

It is capable of producing water pure enough to get reproducibility and accuracies within parts per million when using ion mass spectrometers in analytical geochemistry. Other users of this system include microbiology and atmospheric science


so, how to use the ultra pure MiliQ machine to produce ultra-pure miliQ water?

1st, say the password:MiliQ MiliQ MiliQ MiliQ MiliQ MiliQ MiliQ MiliQ MiliQ

and switch on the power and water input for the machine~


wait a few moments, and press the button and show below


Press this button for water



which the water would come out from this "hose"






OK~~that's all you need to do, but don't forget to turn off the power and water after used everyone

Uninterruptible power supply- in the lab!??

Hello  everyone
i know you guys are having exam, that why i wanna introduce you to this thing below

the
UPS, or so called Uninterruptible power supply
it is basically a battery that would served as a back up when there is a power failure
and therefore the name

the model we using here is a TX600 series, which would hold a power of about 600W.
more than enough to power your laptop for about one whole day

so the front panel only have one button and 3 light, this machine can be turn on by pressing down the button once
and can be turn off by holding it for at least 5 seconds

the light in the middle(green light) means that it is running on AC power
the one on the left(red) indicates an error in current, possibly an overload of current.
the one on the right (orange), means that this thing is running on its battery power
or so called discharging

PS: it will beep very annoyingly every 4 seconds when irunning only on battery power


the back of the ups consist of one socket input( the one on the top). and 4 current output.
It can also serve as a surge protector for your phone and modem

and last but not least, the rotary evaporator pump in action!!!!

Legendary Rotary Evaporator

Hello, i know everyone is quite busy for the exam, 
therefore, 
i plan to update this place much more often so that all of you can actually rest your brain a bit by reading my post
....
OK, maybe not really

anyway, good luck for all of you, hope everyone score HD for everything~!!!

so, as promised, to day i will be explaining the highly sophisticated looking machine 
aka
Rotary Evaporator

so, a rotary evaporator is a device that used to remove solvent from a sample gently and efficiently by applying lower pressure. 

The decrease in pressure also decrease the boiling point of the solvent, which in turn conserve some fragile materials in the extract.

ok, lets start

turn on all this before operation~

so, it consist of (left to right) an air pump, a condenser, and a water bath.


so, this is the air pump that actually sucks air from the condenser

and you will have to fills this tank with water before operation,  this pump was driven by Bernoulli's effect, and the water is used not only to cool down the pump, but also to retain the solvent from escaping into the environment.

now, you connect the tubing to the condenser....

since it was called an rotary evaporator, this thing is used to control the rotation speed of the  thing shown below.

so the rotation prevent the solvent to become superheated
and also helps to create a thin film of solvent across a large surface
this thing was suppose to be connected to the condenser, but since someone is using,
i could only put it into the water bath.~~~~

so this is the collecting flask, which collects your solvent~~



now the water bath sink~it is suppose to be filled with water during operation

PCR Machine-Eppendorf Master Cycler Gradient

Hiiiiiiiiiiiiiiiiiiiiiiiiiiiii Everyone~!!

today, as promise, i will be showing you guys the PCR machine

1st of all, PCR machine(Thermocyle Machine) is always been over rated. Don't be too disappointed once you know exactly how to use it

1st of all, an overview.

and this is to turn on the power, nothing special.

and again turn on the power boringly

now, put in your sample (usually in a 20uL or 50uL tubes), into the bee hive as shown.
 The bee hive is made of metal and can be really hot during operation

now the software, i will not be showing the programing as anyone  who touch this machine will know how to use it.
so, you just have to press enter when the highlight is on the start position.

ok, done...now you just have to wait for roughly 3 hours for the cycle to finish



usually people would run an agarose gel to check for the quality of the PCR,
i would let you know what is that next time

Pollution of water resources in Kuching, Sarawak



Hi, all
As you all know, i walk to school 
and i happen to pass through this "river" every morning

I was totally shocked the first time i saw a river in a blanket of rubbish
Although i don't smell anything rotten inside the river
It was never a good sight to see ~

the location of this river, i have no idea about the exact location as google maps was not updated


OK, now the river, i think is picture is self explanatory enough
this scene you seeing, happens every single time after a heavy rain

Water bottle, trash, polystyrene and anything you can find in a dumping site.

 terrible sight

and another terrible sight.....

Yap, the whole river is contaminated~~

The only thing i wonder is, why no one is taking action on this.
i saw beach cleaning, i saw people going bald for cancer children
i saw changes must be made during the election
i saw article on newspaper saying that how important are our water resources
and i saw people doing pollution studies at remote area
but as always,
