Petri Dish, Balances and tonic water fluoroscences


 HI everyone, hope you have washed your eye balls and prepare yourself for this thrilling ride
today, some simple details about petri dish, electronic balances and quinine will be introduced specially for YOU~~

1st, the most important thing in microbial culture~~~~the PETRI DISH`!!!

A petri dish is 
A shallow, circular dish with a loose cover, usually made of transparent glass or plastic and used to grow cultures of microorganisms. The petri dish is named after German bacteriologist Julius Richard Petri (1852-1921).


the media is usually poured in right after autoclaving (will discuss what is this later, basically a process that could kill every single living thing from earth). The agar is allow to solidify before any inoculations(process of putting the bacteria in ) begins.

 The cap is design to be loose to allow easy access to the interior part, and also too make sure it will not burst during autoclaving. It is usually sealed with para film right after inoculation to avoid any contamination. Then, the whole petri dish is ready to be incubated under desired temperature.

Next, our precious electronic balances. They are pretty expansive compare to those you used in the kitchen for cake making.However, the principle of using remains.

A electronic meter is based on the theory that electric current will be produce by  movement of a magnet within a coil. This lights up a LED and the total light amount is in turn detected by another photosensitive cells. The brightness of the light is then converted into a signal which is known as our mass.

Make sure you always zero the balances before commencing any measurement. How to?
open the switch and wait, it would be done automatically...:)

the total weight of the petri dish show above is 13.77g

Next, i will be introducing how do you actually use a balance to get a precise measurement, 

After zero-ing the instrument, placed your container on top of the balances and record the initial weight.Here, this plastic cup weight is 2.84g

Next, add in the desired amount of solutions and record the final weight. so, the weight of container plus solution would be 26.73


next, measure the container again after the solution is being poured off. therefore the weight of the solution obtain would be?

Now, below is how a nutrient agar was being made using THE balance

The "scott bottle" is placed on THE balances

The powder is added, that is not my hand...i am holding the camera for your interest 

then add in water~~~~~

and go for autoclave~!!!!
thats all we have to do

the solution will then be poured into the petri dish and ready to be used for growing bacteria~~~!!!


Next, our tonic water...due to copyright issue, i cannot tell you the brand~but i think all tonic water should works~~or you can use the ink from highlight pen~~would work better

1st of all, two cups of tonic were prepared,,,this picture is took under normal white light


Next come the UV, they can easily be found in some money examiner

Next, they were poured onto a petri dish for a clearer view of fluorescences~~yap
we would call that a soft glowing
